The Colonial Inn Museum stands on part of an 1834 grant of 995 acres to Mudgee pioneer George Cox which was located on the western boundary of the future town of Mudgee.
In the 1850s, during the gold rush period, George Cox started to subdivide small portions of land on both sides of the road to the north-west which traversed the grant. The museum site was purchased in 1853 by George Brooks who erected the building prior to selling it to Mudgee publican George McQuiggan in 1857.

West End Hotel circa 1910
The building appears purpose-built as a hotel, but in the early 1860s it functioned as a store. The building operated as the West End Hotel until 1923, when it was delicensed; it subsequently became a private house, then a boarding house.
In 1966 the Mudgee Historical Society purchased the building for use as its headquarters and established a museum on site to showcase its splendid collection of Australiana.
